7 glaucoma susceptibility gene regions

Glaucoma is an eye disease with optic nerve damage, which is the first cause of blindness in Japan. The main type is open-angle glaucoma, and the genetic factors of the disease are still poorly known.
The research team published a paper in the British journal of Human Molecular Genetics, saying that they used 3,980 open-angle glaucoma patients and 18,815 control groups collected by the Biobank of Japan, and distributed about 6 million of the entire human genome. Single Nucleotide Polymorphisms (SNPs) were performed for genome-wide association analysis. In addition, SNPs that were identified to be strongly associated with open-angle glaucoma were reproducible by two separate groups (3398 patients, 17570 controls). It was found that seven new gene regions have an effect on the onset of glaucoma. The team used these genetic regions to verify the risk of other races (8,357 patients, 38,100 controls), and identified two SNPs involved in Asian ethnicity and four SNPs involved in European ethnicity. Path analysis of related gene features identified a population of epithelial growth factor receptor signaling-related genes affecting the onset of glaucoma.
The team also evaluated the seven characteristics of the open-angle glaucoma reported in the past and found that open-angle glaucoma has a common genetic background with type 2 diabetes and cardiovascular diseases such as myocardial infarction and ischemic stroke. It means that some people are prone to this disease. They will further study the relationship between systemic status and glaucoma and look forward to contributing to preventive medicine.
