Product introduction:
Connective tissue refers to the three types of fibers contained in the narrow sense: collagen fibers, reticular fibers, and elastic fibers. There are also many methods for connective tissue staining, such as Masson trichrome staining, Van Gieson staining, Gomori ammonia silver method, and Mallory phosphotungstic acid hematoxylin staining. However, the above staining method only focuses on the staining of a certain two tissues.
Russell's modified Movat five-color overdye is popular for its rich, vibrant color. This staining method is mainly used to display atherosclerotic plaques. Weigert hematoxylin is used to stain the nucleus, saffron red is used to stain the cytoplasm, saffron stained collagen tissue, and Alcian blue stained matrix (proteoglycan). Due to the complicated operation process of the kit, the dyeing effect has a great relationship with the operator's experience and the degree of the number, so it is not easy to dye very satisfactory results at the same time.

Bring your own materials:
1, series of ethanol
2, Bouin fixative
3, microwave oven
4, distilled water
Operation steps (for reference only):
1. Paraffin section is routinely dewaxed.
2. Series of ethanol hydration.
3. Take an appropriate amount of Bouin fixative into a microwave oven and heat it for 30 to 60 seconds. Immediately put into a slice for 10 minutes.
4. Rinse with running water for 10 minutes.
5. Slice into the sea wave solution for 5 minutes, rinse with distilled water for 2 to 3 times.
6. Sectioned into Alixin blue staining solution for 20 min.
7. Rinse with running water for 2 to 5 minutes.
8. Warm the oven with a microwave oven. After preheating the alkaline ethanol solution at 45-60 ° C, the slices are placed in an alkaline ethanol solution for 10 minutes.
8. Rinse with running water for 2 to 5 minutes.
9. Slice into pre-formulated reagent (D)-Weigert hematoxylin staining solution and stain for 60 min in the dark.
10. Rinse the running water slightly, rinse with distilled water for 2~3 times, each time for 3~5min.
11. Slice into the pre-formulated reagent (E)-zinc-red magenta staining solution and stain for 1 min in the dark.
12, rinse with distilled water 2 to 3 times, each time 3 to 5 minutes.
13. The slice was processed into a phosphotungstic acid solution for 5 minutes.
14. Directly transfer to a weak acid differentiation solution for 5 min.
15. Rinse the distilled water 2 to 3 times, each time for 3 to 5 minutes.
16. Dehydration: 95% ethanol 1 min, 100% ethanol 2 times, 1 min each time.
17. Slice into saffron staining solution and stain for 5 minutes.
18. Dehydration: Anhydrous ethanol 2 times, 1 min each time.
19. Xylene is transparent and covers the slide.

Precautions:
1. Due to the staining power and tissue section, it is not necessary to display all five colors after dyeing.
2, the thickness of the slice is generally required to be about 5mm.
3. The elastic fiber differentiation is usually completed within 2 to 3 minutes.
4. It is very important to rinse the running water to remove the Weigert hematoxylin staining solution. If the washing fails, the subsequent dyeing step will be inhibited.
5, the operation steps are cumbersome, the dyeing time is often easy to take off the film, pay attention to pre-emptive film treatment.
6. This staining method can display Cryptococcus neoformans and dye it into bright blue.
